How can trispecifics be integrated with CELMoDs in myeloma treatment?

In this multiple myeloma roundtable from EHA 2026, the panel discusses the potential therapeutic combinations of trispecific antibodies and the novel CELMoD mezigdomide in patients with relapsed or refractory multiple myeloma.

The panel was moderated by Paul G. Richardson, MD, clinical director of the Jerome Lipper Multiple Myeloma Center at Dana-Farber Cancer Institute, who speaks with Meletios A. Dimopoulos, MD, professor and chairman of the department of clinical therapeutics at the National and Kapodistrian University of Athens School of Medicine, Rahul Banerjee, MD, FACP, assistant professor in the clinical research division at Fred Hutchinson Cancer Center, and Prof. Katja Weisel, deputy head of oncology, hematology and bone marrow transplantation at the University Medical Center Hamburg-Eppendorf.

The panel discussed combining trispecifics with mezigdomide, noting the strong activity and improved tolerability of targeting BCMA and CD38. They also discussed how mezigdomide might reverse T-cell exhaustion, while cautioning that careful treatment sequencing is necessary because mezigdomide is highly immune-activating.
