Phase 3 SUCCESSOR-2 trial overview

In this multiple myeloma roundtable from EHA 2026, the panel discuss the primary outcomes of the phase 3 SUCCESSOR-2 trial, which evaluated the novel CELMoD mezigdomide in combination with carfilzomib and dexamethasone (MeziKd) versus carfilzomib and dexamethasone alone (Kd) in patients with relapsed or refractory multiple myeloma.

The panel was moderated by Dr. Paul G. Richardson, MD, clinical director of the Jerome Lipper Multiple Myeloma Center at Dana-Farber Cancer Institute, who speaks with Meletios A. Dimopoulos, MD, professor and chairman of the department of clinical therapeutics at the National and Kapodistrian University of Athens School of Medicine, Rahul Banerjee, MD, FACP, assistant professor in the clinical research division at Fred Hutchinson Cancer Center, and Prof. Katja Weisel, deputy head of oncology, hematology and bone marrow transplantation at the University Medical Center Hamburg-Eppendorf.

Dr. Dimopoulos reviewed the clinical efficacy data from the trial and discusses the results, which showed that MeziKd significantly extended progression-free survival in the patients.

The panel goes on to discuss the critical design mechanics and regulatory steps behind the SUCCESSOR-2 trial protocol. They also address the practical logistical advantages that this phase 3 SUCCESSOR-2 trial data offers to healthcare providers across both academic institutions and community practices.
